Optimized Identification of Triacylglycerols in Milk by HPLC-HRMS

Abstract This work has developed an optimized workflow for the targeted analysis of triacylglycerols (TAGs) in milk by liquid chromatography coupled with a Q-Exactive Orbitrap mass spectrometer. First, effects resolution (17,500; 35,000; 70,000; 140,000) and automatic gain control target (AGC, from 2×10 4 , 5 1×10 6 3×10 ) have been goal to minimize injection time, maximize number scans, error. Then, flow rate system was also maximizing theoretical plates. The resulting parameters consisted 200 μL/min, 35,000, AGC . Such optimal conditions were applied TAG fat extracts. Up 14 triglycerides identified performing data-dependent HPLC-HRMS-MS2 experiment (t-SIM-ddMS 2 ). findings reported here can be helpful MS-based lipidomic workflows lipid analysis.
